5Х One-Step Plus RT-PCR Mastermix is a ready-to-use reagent optimized for real-time quantitation of target RNA sequences using hydrolysis probes. In a single tube, RNA is first converted to cDNA by a reverse transcriptase, and then a DNA-dependent DNA polymerase amplifies the cDNA, enabling quantitation via qPCR. 5X One-Step Plus RT-PCR Mastermix contains all the necessary components for RT-PCR assay, including: «warm-start» reverse transcriptase, hot-start Taq polymerase, dNTPs/dUTP and optimized buffer. Genetic modifications of reverse transcriptase provide improved thermostability and increased productivity with GC-rich RNA matrix resulting in the higher cDNA yield. Hot-start modification of DNA polymerase led to significant increase of sensitivity (single copies of RNA) and specificity of the assay. The mastermix also includes thermolabile UDG, which provides protection against carry-over contamination and a human RNase inhibitor to prevent RNA degradation. The presence of the RNase inhibitor allows for the amplification of nasopharyngeal samples stored in Saline and PBS for SARS-CoV2 diagnosis without the need for prior nucleic acid purification. Special additives also increase resistance to inhibitors.
