Description
Thermolabile Uracil-DNA Glycosylase (tUDG) is a 25.5 KDa protein isolated from a recombinant E. coli strain bearing cloned UNDG gene from a psychrophilic marine bacterium. It hydrolyzes uracil-glycosidic bonds in single- or double-stranded DNA, creating alkali-sensitive abasic sites. These abasic sites can be hydrolyzed by endonuclease, heat, or alkali treatment. The enzyme is used in the preparation of PCR, RT-PCR and LAMP mixtures to prevent carryover contamination.
The enzyme is highly purified by affinity and anion-exchange chromatography. The enzyme is inactivated completely during 5 minutes at 55°С and does not interfere with the current reaction amplification products.
